Today I checked the tadpoles I injected and electroporated on Tuesday to see
if I had any transfection. To determine whether or not my GCAMP plasmid DNA
just had poor fluorescence, I had used John's NFAT plasmid DNA to inject in
the tadpoles on Tuesday. Two of the tadpoles did have some decent
transfection when I checked them under the wide-field fluorescent
microscope! So, we are not quite sure if the problem was due to the DNA or
due to electroporation technique and/or voltage. At any rate, I look
forward to familiarizing myself with the confocal microscope tomorrow to get
3-D images of the successfully transfected neurons! Our next conundrum to
solve is how to get the GCAMP plasmid DNA to successfully transfect so that
we can then determine the calcium ion flow.

Regarding the bacterial culture mystery. . . . apparently 3 times is not a
charm. The 3rd culture we began last night still did not grow. Dr. M
suspects that an improperly prepared LB broth may be the culprit. He is
planning on requesting a new batch be made before we start our next culture
and then we shall see. . . Stay tuned!
