Today I stained four different slides from the two different kinds that I had prepared yesterday. I altered a different variable as well. In the past we have been concentrating on the wash conditions as our variable, but with these four I experimented with the 2-Iminothiolane concentration. I stained two with just antibodies, and the remaining two with antibodies and bacteria. Once the slides were prepared and stained, I viewed them under the microscope and took five pictures of each spot and the middle. I then imported these pictures into Image J where I was able to use a Macro to quantify the bacteria and clusters on each of the slides and spots. I then plugged this data into my spreadsheet and compared it to the existing data. Unfortunately no real changes occurred. We still have very limited specificity. I also made more coupling buffer (Again!) and worked on my power point.
