Today I added sulfhydryl groups to the antibody for coupling, cross-linked the sulfhydryl containing antibody to the glass slide, stained one of my slides, and then viewed it under the wide field fluorescent microscope. My antibodies stuck and I got two great pictures from the microscope. One each of the salmonella antibody and the E. coli antibody. I also checked on the bacteria that I started growing in preparation for tomorrow. My E. coli is growing beautifully, the salmonella hasn't done much of anything yet. So, it looks like tomorrow will be strictly an E. coli day.
