Today I added sulfhydryl groups to the antibody to prepare it for coupling to the glass slide using 2-Iminothiolane * HCL (Traut's Reagent). Next, I removed the excess Iminothiolane using a desalting column. After that, I treated the slides with a cysteine solution to prevent the maleamide groups from reacting. Finally, I cross-linked the sulfhydryl containing antibody to the activated glass surface. The slides were allowed to incubate for 4 hours and then were rinsed with coupling buffer. Once they were dry, I stored them in the refrigerator in preparation for staining tomorrow. I hope to have a picture from the microscope proving the existence of my antibody by the end of the week.
