This morning Dr. Miskevich and I wrote out 7 different procedures, each altering a different variable because our initial trial failed yesterday. We also noticed that a baby chick had begun the hatching process and I named him Pete. This afternoon I implemented the 7 different procecdures onto 7 different slides. This was extremely stressful, as I was doing them in parallel and was constantly dashing about the lab to rinse stuff, move stuff, and incubate stuff. Once the slides were successfully prepared, I took them to the microscope room and looked for any noticeable changes in consistency. There were none. So I took 5 pictures of each spot (two on each slide) for all 7 slides under the microscope. Tomorrow I will use these pictures to use Joseph's project so that we can quantify the bacteria and come up with some hard numbers that may aid us in our search for the right coupling/staining procedure. We have a few more variables to try also, so that will either be done tomorrow or Monday. Oh, and Pete didn't make much progress coming out of his shell, but I'm taking him home with me when he hatches.
