This morning I practiced the injection and transvection procedure some more.
After lunch, John (the graduate assistant) showed me the fluorescent
microscope. We secured the tadpoles by placing them in a small groove
carved into the bottom of a petri dish filled with water. A weight
constructed of a bent nail with fishing wire tightly strung across it served
to anchor them into the groove so they would remain stationary. After
learning how to use the microscope and observing the fluorescence of the
transfected cells, I practiced some more at extracting the brain from
several specimens. At the end of the day, Dr. Miskevich helped me begin the
culture needed to prepare our DNA. Once it was prepared, I placed it in the
incubator for overnight culturing.
