Today I made a spreadsheet from the data that I collected from my slides on Friday. On the spreadsheet, I calculated the average number of bacteria on each spot on each slide and the standard deviation between pictures. This information enabled us to narrow down the procedure with the most positive results. I then repeated this procedure on my last remaining prepared slide to try and replicate the results. Once the slide was prepared, I viewed it under the microscope, took pictures of each spot, and quantified them using the Image J bacterial analysis macro. These numbers were then plugged into the spreadsheet along with the others. We were excited to note that our results were very similar to the previous identical trial. Thus, this method, while still producing rudimentary results, is definitely producing positive results. After that I made more coupling buffer and 1x PBS solution because I had run out. I also prepared 20 slides by maleimide activating them, and then coupling the antibodies to the glass. This process definitely took the longest because I crammed two days worth of work into one with this process alone.
